That isn't going to force it to use a specific tuner. That'll just cause the tuner to sit there, waiting for the signal.
At the very least you'd need to disable the tuner don't want it using. You do this in NextPVR's Settings page. Hold the Ctrl key and you'll get the advanced options for disable/delete. You'd need to disable the one you don't want it to use (and possibly restart the server).
Realistically, you might be better off playing around when you've got the splitter and signal feed to both. (though splitters with DVB-S can be tricky, since often they want power supplied by one of the inputs with power pass)
